Dont take it off :L You'll find lots of brake fluid and the piston. I just nipped mine up (it was quite loose) tested it, and boom, leak free And what other people have said about the humidity in your basement could have an effect, but i cant see humidity causing your brake to be crap for the first hour of a ride.

1000psi Its personal preference. Some people run higher pressures, some run lower. What I do is pump it up to 30psi, then let it down till it feels right.
